As above.
I always mask the area off, not for a neater cut, but so you can mark the measurements easy.
After the first hole is done I always check that my 2nd measurement hasn't slightly changed by standing the tap in place.

A couple or four of tips -
[1] measure twice and cut once, if in doubt measure again
[2] Always drill a small pilot hole in Acrylic first
[3] Always use the smallest hole saw you can for the o/d of the tap threads - ie if you need a 22mm hole, don't use a 25mm as its all you have, go out and get a 22mm
[4] Always use sharp drills and sharp bimetal hole saws, blunt old tatty drill and saws will burn and slip laterally and create an elipitical hole.
