No no no no no....... Plasterboard adhesive is gypsum based and relies on its mechanical bond with the substrate which in turn relies on its ability to key to the substrate. Paint will very significantly reduce the bond strength and could easily lead to a failure. If you cannot remove the paint use mechanical fixing instead.
