I don't use ready mixed adhesive so I wouldn't know about that. What I do know is, if Done as I said for prep and use powdered adhesive, the next day you cannot pull the tile off the wall.
Don't ever tile on top of paint on a plastered wall, if it's a **** coat then it probably won't come off but any more layers then it must be removed.
Don't just score the surface to get a key as its only minimal contact, remove as much paint as possible & you will be good to go.

The use of tubbed adhesive(because it is cheap) to remove the paint is a tried and tested method of removing new paint on new plaster,but powdered adhesive has the same desired effect........ie paint will affected by moisture.....be it whether it has a tile placed on it or not.
As for not being able to pull the tile off a wall the next day.......I have been able to take a tile fixed with RS onto a plastered wall the next day quite easily.......even RS doesn't cure fully for 4 weeks.

I agree it is always best to get the paint off! I wouldn't tile on any layers of paint or any bathroom paint or anything like that. Only a sealing coat of thin white emulsion.
