Yes, you can take your measurement from the plinths to get your room centre if you can't do it from the walls. Are you planning to tile up to the plinths? You are always better to remove your plinths, tile under the units slightly & cut your plinths to suit. Leaves a much neater finish and makes removal of your plinths far easier in the future
