you will have to incorparate an expansion joint between the two substrates ply and screed and carry it threw the tiles, especially as there is under floor heating as the two substrates expand at different rates

as you have got ply butting up aginst screed the two different substrates can expand at different rates to each other, this will put pressure against where the two substrates meet. there for you need to stop this from happening, if you put in an expansion joint between the two different substrates then this will counter act that affect.
You still need for the expansion joint to carry on up threw the tiles so as this movement is carried threw the tiles if NOT then the stress of this movement will crack the tiles.

Couldn't have put it better myself peckers......I would probably have taken 3 pages to do it as well.

Other thing to consider is whether the concrete is dry. I assume it has been down for some time and is not a new build with a new slab.
